摘 要:目的评价自行研制的Comfort-DAⅡ型高分子合成树脂类义齿粘附剂的全身毒性及细胞毒性。方法采用经口途径全身急性毒性实验;细胞毒性实验中将Comfort—DAⅡ置于细胞培养液72h制备出材料的浸提液,用同一培养液再稀释为原浓度的50%、75%后分别加入含人口腔成纤维细胞悬浮液的培养板中,其中空白对照组仅加入新鲜培养液。3块培养板分别继续在5%二氧化碳、37℃培养箱内开放培养2、3、4d后,四唑盐比色法观察细胞的活性以研究细胞增殖率,用分光光度计以490nm波长测光密度值(optical density,OD),组间方差分析并评定该材料的毒性级。结果未见任何急性毒性反应;空白对照组、Comfort-DAⅡ的50%和75%浓度组间OD值的差别有统计学意义(P〈0.05);2组浓度Comfort—DAⅡ的毒性程度为“0~2”级。结论所研制的义齿粘附剂对机体无毒性。Objective To evaluate the toxicity of synthetic maeromolecular resin class of Comfort-DAⅡ denture adhesives, which was developed by the authors. Methods Acute toxicity was tested with standard method. While the tyrotoxicity of Comfort-DA was tested as the following methods. A sample of Comfort-DAⅡ was prepared and extracted in culturing medium to prepare the eluates. Then the eluates were diluted by culturing medium as 50% and 75% concentration to be assessed for cytotoxicity on HOFs by means of tetrazolium bromide (MTT) colorimetric assay. Wells containing fresh medium alone served as controls. Cell viability was recorded by optical density after culturing in an atmosphere of 5% CO2 and 95% air at 37 ℃ for 2, 3 and 4 days respectively. The viability of HOF ceils was evaluated by the MTT assay to investigate cell proliferation. Optical density (OD) was measured by a speetrophotometer at 490nm. Then evaluating the cytotoxieity grade in test groups according to the means of cell proliferation. ANOVA was used to test for statistical significance. Results No toxicity effect was observed with in vivo test. While the statistical analysis of results of the MTT cytological assay indicated there was significant difference (P 〈 0.05 ) in OD (indicate cell viability) between all concentrations of Comfort-DA and the controls at all incubation times. The results of cell proliferation percentage also showed that the eytotoxicity grade of tested material only displayed "0 -2". Conclusion The developed demure adhesive did not exhibit toxicity.
