摘  要:目的:观察逆转录酶抑制剂叠氮胸苷(齐多夫定,Azidothymidine,AZT)联合放射线对人喉鳞癌Hep-2细胞裸鼠皮下移植瘤的治疗作用。方法:建立人喉鳞癌荷瘤裸鼠模型,应用AZT联合放射线治疗人喉鳞癌裸鼠移植瘤,观察其对肿瘤体积、端粒酶活性、hTERT蛋白表达效率及凋亡的影响。结果:与对照组相比,放射组和AZT+放射组肿瘤生长速度受到明显抑制(P<0.05),提示放疗、AZT联合放疗均能抑制肿瘤生长,且后者抑瘤率显著高于前者(P<0.05)。端粒酶活性(TA)及hTERT蛋白检测结果一致,即放射组>对照组>AZT+放射组>AZT组,提示低剂量放疗能够引起肿瘤细胞端粒酶活性的升高,而AZT能够有效抑制抑制辐射所致的端粒酶活性增高。凋亡指数为AZT+放射组(9.60±0.97%)>单纯放射组(6.91±1.65%)>AZT组(5.17±0.71%)>对照组(0.77±0.27%)。结论:AZT联合放射线能有效的抑制人喉癌裸鼠移植瘤的生长,AZT能增加肿瘤的放射敏感性。Objective: To observe the effect of the telomerase inhibitor, Azidothymidne (AZT), combined with radiotherapy on the xenograft tumor of human laryngeal squamous carcinoma in vivo. Methods: Xenograft tumor models of human laryngeal squamous carcinoma were established. The AZT and radiotherapy were used to treat the xenograft tumor of human laryngeal squamous carcinoma for evaluating their influence on the tumorigenicity, the telomerase activity (TA), the expression level of hTERT protein and the apoptosis index (AI). Results: Compared to that of the control group, the growth velocity of tumors of radiation group and AZT plus radiation group was obviously suppressed. It indicated that both radiation and AZT plus radiation could effectively inhibit the growth of tumor, and the latter had significantly higher effects than the former did (P〈0.05). The results of TA were in accordance with the expression of hTERT protein as: radiation treated group 〉 control group〉AZT plus radiation group〉AZT group. The apoptosisindexes from the highest to the lowest were AZT plus radiation group (9.60±0.97%)〉radiation group (6. 91± 1. 65%)〉AZT group (5. 17±0. 71%)〉control group (0. 77±0. 27%). Conclusion. AZT combined with radiation can effectively inhibit the growth of Xenograft tumor of human laryngeal squamous carcinoma in nude mouse. AZT can increase the radiation sensitivity of tumors.
