摘  要:目的:探讨环孢素A(CsA)对人滋养细胞系Bewo的23号非转移性基因(nometastatic gene 23)H1型即nm23-H1表达的调控作用,为治疗滋养细胞疾病提供新的依据。方法:将Bewo细胞分为对照组(加溶媒)、环孢素组加入终浓度由10-2μmol/L-10μmol/L环孢素A。分别于48h后用RT-PCR检测nm23-H1基因mR-NA表达水平,于72h后用Incell Western分析nm23-H1蛋白表达水平。结果:与对照组相比,nm23-H1的表达水平随环孢素A浓度10-2μmol/L-1μmol/L呈现降低趋势,其中1.0μmol/L环孢素Anm23-H1 mRNA和蛋白水平均明显降低(P<0.05,P<0.01);当浓度升高至10μmol/L时,nm23-H1则呈升高趋势。结论:低浓度环孢素A可通过降调节nm23-H1的表达,继而改善滋养细胞的侵袭力。AIM : To explore whether cyclosporine A (CsA) can regulate the expression of nometastatic gene23 H1 type ( nm23 - H1 ) in human choriocarcinoma Bewo cells, in order to seek new proof of treating trophoblast diseases. METHODS: The Bewo cells were divided into two groups. The vehicle control group, and the CsA group with different concentrations from 10 -2 μmol/L to 10 μmol/L. The effect of CsA on the transcription of nm23 - H1 was determined by reverse transcription - polymcrase chain reaction ( RT - PCR) after cultured for 48 h and protein level of nm23 - H1 was determined by In - cell Western after cultured for 72 h. RESULTS : Compared to the vehicle group, CsA significantly down- regulated the transcription and translation of nm23 -H1 in a dose -dependent manner from 10-2 μmol/L to 10 μmol/L, and the inhibition reached its top when concentration of CsA was 1.0μmol/L (P 〈 0. 05, P 〈 0. 01 ). However, when concentration of CsA rised to 10 μmol/L, the expression of rim23 -H1 increased significantly. CONCLUSION: The result above indicates that CsA at an appropriate lower concentration can inhibit the expression of the nm23 - H1, which involves in invasiveness of human trophoblast cells.
