摘 要:目的:探讨Sonic hedgehog信号通路中锌指转录因子Gli1在胰腺癌组织中的表达及其意义。方法:收集56例手术切除的新鲜胰腺癌及癌旁胰腺组织,应用免疫组化S-P法及RT-PCR方法检测癌组织和癌旁胰腺组织Gli1蛋白及Gli1 mRNA的表达情况。结果:Gli1蛋白和Gli1 mRNA在癌旁胰腺组织中不表达或弱表达,在胰腺癌组织中的表达分别为85.7%和73.2%,两者在胰腺癌组织和癌旁胰腺组织中表达率差异有显著性。Gli1蛋白和Gli1 mRNA阳性表达率与胰腺癌的分化程度有关,与肿瘤大小及有无淋巴结转移无关。结论:Gli1的异常表达与胰腺癌的发生发展密切相关,针对的干预治疗有可能成为治疗胰腺癌的一种新的策略。Objective: To study the expression and significance of zinc finger transcription factor Gli1 mRNA in pancreatic carcinoma. Methods: Tissues of 56 pancreatic carcinoma and latero-carcinoma were assembled,the expression of Gli1 proteins and Gli1mRNA were detected by immunohistochemistry and RT- PCR. Results: Only weak expression, if any, of Gli1 proteins and mRNA were detected in latero-carcinoma tissues, but in pancreatic carcinoma,the expression of Gli1 proteins and mRNA were 85. 7% and 73. 2% respectively, there was significant difference between pancreatic carcinoma and latero-carcinoma tissues, the expressions of them were concerned with differentiation of cancer , not tumorous size and lymphonodulus transfusion. Conclusion: Gli1 expression is correlated with pancreatic tumorigenesis closely, and could be served as a potential therapeutic target.
