摘 要:目的观察骨髓间充质干细胞(BMSC)移植对脑缺血皮质区血管形成的影响及微血管Notch1表达的变化。方法用线栓法复制大鼠脑缺血再灌注损伤动物模型,随机分为假手术对照组、单纯脑缺血损伤组和脑缺血后BMSC移植组。采用免疫组织荧光染色法检测各组缺血皮质区Ⅷ因子及Notch1的表达。结果(1)植入的BMSC可以趋向性迁移到脑缺血损伤区域。(2)BMSC移植组脑缺血皮质区新生微血管密度较单纯脑缺血损伤组和假手术对照组明显增高。(3)BMSC移植组缺血皮质区多数微血管内皮细胞阳性表达Notch1,且明显多于单纯缺血组和假手术对照组。结论BMSC移植可促进脑缺血区血管新生,且可能与促进Notch1表达有关。Objective To observe the effects of bone marrow mesenchymal stem cells transplantation on angiogenesis and Notchl expression in cerebral isehemic area. MethOds Middle cerebral artery occlusion models were made by intraluminal thread method. Animals were divided into three groups randomly (cerebral ischemia group, BMSC transplantation group and sham operation control group). Immunofluorescence methods were used to investigate the expressions of Factor Ⅷ and Notch1 in ischemic cortical areas. Results (1) The transplanted BMSC can move to cerebral ischemic regions. (2)The microvessel density in the injured cortical areas treated with BMSC were significantly higher than those of the cerebral ischemia group and sham operation control group. (3)The coexpression of Ⅷ and Notchl in injured cortical areas treated with BMSC were significantly higher than those of cerebral ischemia group and sham operation control group. Conclusion These results suggest that BMSC transplantation can improve the microvessel density in the injured cortical areas by promoting the expression of Notchl.
