脱细胞角膜基质的制备及其超微结构

摘　　要：目的探讨异种角膜脱细胞基质的制备方法,观察异种脱细胞角膜基质的超微结构。方法取新鲜狗角膜进行脱细胞处理,用光学显微镜和扫描电镜观察植片的形态学改变。从一脱细胞狗角膜基质取1mm厚组织片,植入兔角膜囊袋中,分别于术后不同时间点对其进行临床观察及组织切片看形态学改变,及其与受体组织融合情况。结果脱细胞后的狗角膜组织呈现无细胞的三维网状,肉眼观察为灰白色半透明组织,经脱水处理后呈透明状,组织切片中未见蓝染的细胞核结构,扫描电镜示基质纤维完好,呈有序的平行结构排列的透明结构。植入囊袋中可见局部水肿不明显,呈半透明状,术后1个月可见有新生血管长入。结论脱细胞处理方法可以有效的脱净细胞,并保持角膜三维结构和透明性,宿主细胞可以长入异种脱细胞角膜基质植片中并在植片中增殖,与其他方法处理的脱细胞基质相比,组织的生物相容性更好。[Objective] To examine corneal ultrastructure of xenogenic decellularized corneal matrix. [Methods] Fresh dog corneas were decellularized by serial digestion confirmed by light microscope （LM） and transmission electron microscope （TEM）. The decellularized corneal piece was implanted through a mid-depth circumferential incision into 8 rabbit corneal stroma. Corneal opacity and neovascularization were evaluated postoperatively by slit lamp, and stromal ultrastructure was observed by scanning electron microscopy and transmission electron microscopy. [Resuits] There was no any cell detected after the decellularized procedure. They remain the perfect three dimensional reticular formations. The decellurized corneal piece was ivory white in gross and turned to be transparent after the dehydration procedure. The decellularized corneal piece merged into the rabbit corneal, but new vessel could be seen 1 month postoperatively.[Conclusion] The decellularized corneal matrix can be promising corneal substitute for corneal transplantation because of its transparency and perfect ultrastructure similar to the original corneal.
