摘  要:[目的]探讨表没食子儿茶素没食子酸酯(EGCG)对人肝细胞(L02)损伤的保护作用。[方法]通过测定细胞存活率及细胞培养上清液中转氨酶(ALT、AST)、乳酸脱氢酶(LDH)、MDA含量及SOD活性,研究EGCG对L02损伤的保护作用。[结果]与对照组相比,肝细胞损伤模型组的SOD活性显著降低,MDA含量极显著升高,EGCG各剂量组均可显著降低MDA含量,其培养上清液中ALT和LDH含量显著升高。与模型组相比,EGCG 1.25×10-5、2.50×10-5、5.00×10-5和1.00×10-4mol/L剂量组的细胞培养上清液中ALT含量显著降低,且EGCG 2.50×10-5、5.00×10-5和1.00×10-4mol/L剂量组的细胞培养上清液中LDH含量显著降低;EGCG 1.25×10-5和2.50×10-5mol/L剂量组可使SOD活性极显著增加,EGCG 5.00×10-5和1.00×10-4mol/L剂量组可极显著降低MDA含量。[结论]EGCG可减轻CC l4及H2O2所致的肝细胞氧化损伤,对肝细胞具有保护作用,并具剂量依赖性。[ Objective ] The aim was to discuss the protection of epigalloeatechin-3-gallate (EGCG) on human liver ceils (L02) from injury. [ Method] Through determining cell survival rate, the contents of transaminases ( ALT and AST), lactate dehydrogenase (LDH) and MDA and the activity of SOD in the cell culture supernatant, the protection of EGCG on L02 from injury was researched. [ Result] Compared with the controlled group, in the model group with liver cell injury, the activity of SOD was decreased significantly, the content of MDA was increased extremely significantly, all the close groups of EGCG could significantly decrease the content of MDA and the contents of ALT and LDH were increased significantly in its culture supernatant. Compared with the model group, the contents of ALT were decreased significantly in the cell culture supernatants of the dose groups 1.25 × 10^ -5, 2.50 × 10^ -5, 5.00× 10^ -5 and 1.00 ×10 ^-4 mol/L of EGCG and the contents of LDH were decreased significantly in the cell culture supernatants of the close groups 2.50 × 10^ -5, 5.00 × 10 ^-5 and 1.00 × 10 ^-4 mol/L of EGCG; the dose groups 1.25 × 10 ^-5 and 2.50 × 10 ^-5 mol/L of EGCG could extremely significantly increase the activity of SOD and the close groups 5.00 × 10^-5 and 1.00 × 10^-4 mol/L of EGCG could extremely significantly decrease the content of MDA. [ Conclusion] EGCG could reduce the oxidative injury of liver cells caused by CC14 and H2O2. It had protective effect on liver cells in a dose-dependent manner.
