摘 要:目的观察人肺腺癌细胞株A549及其耐顺铂细胞株(A549/DDP)对X射线的不同反应,不同放射剂量、照射时间对细胞周期的影响。方法用Vi-Cell细胞活力分析仪绘制A549及A549/DDP生长曲线;分别用4、8、12、16Gy6MVX射线照射A549及A549/DDP,以流式细胞仪检测细胞周期变化。结果A549与A549/DDP存活率分别为83.12%±2.23%,92.00%±2.31%(P<0.05)。与亲本细胞株相比,A549/DDPG0/G1期比例降低(P<0.01),G2/M期比例升高(P<0.01)。X射线照射后两种细胞出现G2/M期阻滞,照射后24h达到最大阻滞高峰,48h显示最大峰值回落。G2/M期阻滞强度在24h时与放射剂量呈正相关。在4Gy照射后,A549在6h即出现G2/M期阻滞,但在照射后12h阻滞解除;而A549/DDP在24h内G2/M期阻滞呈递增趋势。在16Gy照射后,A549及A549/DDP6~12hG2/M期阻滞迅速增加,并持续至24h。结论A549/DDP较其亲本细胞株存活能力更强,对于放射线的敏感性增加。X射线可以改变A549及A549/DDP的细胞周期进程。A549经高放射剂量照射后细胞周期阻滞明显,而A549/DDP无论经过高放射剂量还是低放射剂量照射均表现出明显G2/M期阻滞。Objective To investigate the different influence of X-ray on human lung adenoid cancer cell lines (A549) and its cisplatin-resistant cell line(A549/DDP) and observe the cell cycles at different dosage and irradiation time . Methods The Vicell series cell viability analyzer was applied to diagraph the growth curve of them. The A549 and A549/DDP were irradiated by X-ray at different dosage. Cell cycles were checked after the irradiation with flow cytometry. Results The survival rate of A549 and A549/DDP were 83.12 % ±2.23 % and 92.00 % ± 2.31%(P 〈 0.05), respectively. The G0/G1 percentage of A549/DDP was lower than that of A549, but the GJM percentage was higher (P 〈 0.01). With different single dosage of irradiation, X-ray caused A549 and A549/DDP accumulated in G2/M phase. The G2/M arrest was released at 48 h after irradiation. The G2/M arrest was correlated with the radiation dosage at 24 h. At 4 Gy, the G2/M arrest of A549 appeared at 6 h after irradiation but released at 12 h, while A549/DDP was arrested at G2/M until 24 h. At 16 Gy, the G2/M arrest of A549 and A549/DDP were existed. Conclusion The viability of A549/DDP is more stronger than its parental cell line, and it is more sensitive to irradiation. X-ray caused cell lines accumulated in G2/M phase. The significant arrest of G2/M phase is found at high dosage irradiation in A549 but it is showed in A549/DDP whether at high or low irradiation dosage.
