摘 要:目的探讨artemin、GFRα3在胰腺癌中表达的意义及其与胰腺癌神经浸润转移的相关性。方法应用免疫组化链酶菌抗生素蛋白-过氧化物酶法和RT-PCR检测胰腺癌组织、癌旁组织和正常胰腺组织中artemin、GFRα3的表达,观察artemin、GFRα3的阳性表达率与胰腺癌神经浸润转移之间的相关性。结果胰腺癌组织中artemin和GFRα3阳性表达率分别为72.09%和67.44%,均显著高于癌旁组织中表达水平(18.19%,22.73%)。Artemin、GFRα3在有神经浸润的胰腺癌组织中阳性表达率均明显高于无神经浸润癌组织(χ2=11.11,11.78,P<0.01),胰腺癌组织artemin mRNA表达量(0.741±0.014)明显高于正常组织(0.101±0.031)(P<0.05),有神经浸润癌组织artemin mRNA表达量(0.843±0.012)明显高于无神经浸润癌组织(0.512±0.017)(P<0.05)。结论Artemin、GFRα3的表达在胰腺癌神经浸润转移过程中起着重要作用,可能为反应胰腺癌生物学行为的重要指标。Objective To investigate the association of artemin and GFRα3 expressions with perineural invasion and metastasis of pancreatic carcinoma. Methods Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry were used to detect the expression of artemin and GFRα3 in pancreatic carcinoma tissues, adjacent tissues and normal pancreas tissues, and therelevance of artemin and GFRα3 expressions to the perineural invasion and metastasis of pancreatic carcinoma were analyzed. Results The positivity rates of artemin and GFRα3 expressions were 72.09% and 67.44% in pancreatic carcinoma, respectively, significantly higher than those in the adjacent tissue (18.19% and 22.73%). The positivity rates of artemin and GFRα3 expressions were significantly higher in patients with perineural invasion than in those without perineural invasion (χ^2=11.11 and 11.78, respectively, P〈0.01). Significantly higher expression of artemin mRNA was noted in pancreatic carcinoma (0.741 ±0.014) than in the normal pancreas tissue (0.101 ±0.031, P〈0.05), and patients with perineural invasion showed significantly higher positivity rates of artemin mRNA expression (0.843±0.012) than those without perineural invasion (0.512±0.017, P〈0.05). Conclusion Artemin and GFRα3 expressions may play an important role in perineural invasion of pancreatic carcinoma and can be used a useful indicators for evaluating the biological behavior of pancreatic carcinomas.
