摘 要:目的探讨白藜芦醇对重症急性胰腺炎(SAP)大鼠腺泡细胞凋亡的影响及其机制。方法以3.5%牛磺胆酸钠诱导SD大鼠SAP模型,采用全自动生化分析仪检测血清淀粉酶浓度,应用TUNEL染色检测胰腺腺泡细胞凋亡,RT-PCR及Western blot技术检测胰腺组织凋亡调控基因Fas和FasL mRNA及蛋白的表达,以及白藜芦醇治疗对腺泡细胞凋亡及Fas和FasL表达的影响,光镜下观察胰腺组织的病理变化。结果与SAP组相比,白藜芦醇治疗组血清淀粉酶水平及胰腺病理损害评分明显下降,腺泡细胞凋亡指数明显升高,FasL mRNA及其蛋白的表达水平亦显著升高。结论白藜芦醇通过上调胰腺组织凋亡调控基因FasL的表达诱导损伤的胰腺细胞凋亡以减轻胰腺组织病理损害,可能对SAP具有重要的治疗作用。Objective To investigate the effect of resveratrol on the apoptosis of pancreatic acinar ceils in rats with severe acute pancreatitis (SAP) and explore the mechanism of such effect. Method SD rats with 3.5% sodium taurocholate-induced SAP were treated with resveratrol, and the serum amylase was detected with automatic biochemistry analyzer. The apoptosis of the pancreatic acinar cells in the rats was detected by TUNEL assay, and the expression of Fas and FasL genes was determined by RT-PCR and Western blotting. The pathological changes of the pancreas were observed under optical microscope. Results Compared with SAP group, the resveratrol-treated rats showed obviously decreased serum amylase and scores for pancreatic histopathological lesions. Resveratrol treatment significantly increased the apoptotic indices of pancreatic acinar cells and the levels ofFasL mRNA and protein in rats with SAP. Conclusion Resveratrol produces important therapeutic effect on SAP in rats by inducing pancreatic acinar cell apoptosis possibly as a result ofup-regnlated FasL gene expression.
