摘　　要：目的观察法舒地尔对慢性脑缺血大鼠学习记忆及皮层Caspase-3、Bcl-xl表达的影响。方法采用双侧颈总动脉永久性阻断法(2VO)建立慢性脑缺血模型,应用Morris水迷宫检测学习记忆能力,免疫组化方法检测凋亡调控基因Caspase-3、Bcl-xl表达。结果2VO组和法舒地尔组大鼠与假手术组相比逃避潜伏期和游泳路径均明显延长,法舒地尔组学习记忆成绩均明显优于2VO组(P<0.05),缺血3周起,缺血组大鼠皮层Caspase-3表达明显增高,Bcl-xl表达明显降低,法舒地尔组Caspase-3表达下降,Bcl-xl表达增加,与2VO组相比差异显著(P<0.05)。结论法舒地尔能可能通过影响细胞凋亡调控基因变化减轻神经功能损伤而改善学习记忆功能。Objective To investigate the protective effect of fasudil to rats with learning and memory deficit due to chronic cerebral ischemia by mediating expression of Caspase-3 and Bcl-xl gene. Methods Animal models are established by 2VO method, learning and memory function tested with Morris water maze, Caspase-3 and Bcl-xl measured with method of immunohistochemistry. Results The latency in each trial and swim distance of the 2VO and Fasudil group are longer than the sham operation. Expression of Caspase-3 increased and Bcl-xl decreased in the 2VO group, vice versa in the fasudil group. Conclusion Fasudil have protective effect and can improve learning and memory function of 2VO rat by mediating apoptosis gene expression.
