血管抑素在前列腺癌中的表达及与VEGF相关性的实验研究

摘　　要：目的:探讨血管抑素(AS)和血管内皮生长因子(VEGF)在前列腺癌和前列腺增生疾病中的表达及其相关性。方法:选用前列腺癌和前列腺增生术后组织标本各30例,采用免疫组织化学和western blotting方法,检测前列腺癌组织中AS和VEGF蛋白含量的变化情况。结果:在前列腺癌中血管内皮生长因子的平均灰度值与前列腺增生组织相比呈显著增高,而血管抑素则比前列腺增生组织呈显著降低;而且在前列腺癌组织中血管抑素与血管内皮生长因子之间呈显著负相关(r=-0.560P<0.05)。结论:在前列腺癌组织中血管抑素表达量较前列腺增生组织减少,而血管内皮生长因子表达量较前列腺增生组织呈明显增加趋势,两者间呈显著负相关;血管抑素与血管内皮生长因子可能在前列腺癌组织的发生及生长机制过程中发挥一定的生物学作用。Objective： To observe the relationship between VEGF and angiostatin in patients with carcinoma of prostate and benign hyperplasia of prostate. Methods ： 30 carcinoma of prostate tissue and 30 benign hyperplasia of prostate tissue were tested by using immunohistochemical and Western blotting technology. Results： The positive staining average grey value of angiostatin in carcinoma of prostate was increased significantly compared with that of benign hyperplasia of prostate, hut the positive staining average grey value of VEGF in carcinoma of prostate was decreased compared with that of benign hyperplasia of prostate. The negative correlation was found between A S and VEGF （ r = - 0. 560 P 〈 0.05 ; ）. Conclusion： The level of angiostatin expression is reduced in carcinoma of prostate; however , the level of VEGF expression is increased significantly in carcinoma of prostate, and the negative correlation was found between AS and VEGF in carcinoma of prostate）. AS and VEGF may effect development and growth mechanism of carcinoma of prostate in biology.
