二甲基亚硝胺和部分肝切除术对肝脏DNA合成的影响

摘　　要：目的：探讨二甲基亚硝胺（Ｎ－ｎｉｔｒｏｓｏｄｉｍｅｔｈｙｌａｍｉｎｅ，ＮＤＭＡ）和部分肝切除术（ｐａｒｔｉａｌｈｅｐａｔｅｃｔｏｍｙ，ＰＨ）对大鼠肝脏ＤＮＡ合成的影响。方法：用免疫组织化学双ＰＡＰ法检测大鼠肝组织５′－溴脱氧尿嘧啶（５′－ｂｒｏｍｏｄｅｏｘｙｕｒｉｄｉｎｅ，ＢｒｄＵ）标记细胞。结果：１次或４次ＮＤＭＡ作用后５小时至２周，明显可见ＢｒｄＵ标记的肝细胞和肝窦壁细胞，四次ＮＤＭＡ处理ＢｒｄＵ标记细胞数高于一次处理。ＰＨ加一次ＮＤＭＡ处理后５小时至１周内，两类细胞ＢｒｄＵ标记细胞数高于相应单纯ＮＤＭＡ处理。ＰＨ加四次ＮＤＭＡ处理后５小时，ＢｒｄＵ标记肝细胞数高于相应单纯ＮＤＭＡ处理。ＢｒｄＵ标记细胞主要分布在门管区周围。结论：ＮＤＭＡ可刺激肝细胞和肝窦壁细胞ＤＮＡ合成，ＰＨ和ＮＤＭＡ对肝细胞和肝窦壁细胞的ＤＮＡ合成有协同刺激作用。bjective: To determine the effect of Nnitrosodimethylamine(NDMA) and partial hepatectomy(PH) on DNA synthesis in the liver of rats.Methods: Immunohistochemical double PAP technique was used for detecting 5′bromodeoxyuridine(BrdU) to quantify the DNA synthesis.Results: BrdUlabelled hepatocytes and hepatic sinusoidal wall cells were significantly found from 5 hours to 2 weeks after treatment of a single or four times of NDMA,however,the numbers of BrdU positive cells of liver was higher in the four times of NDMAtreated animals than in the a single NDMAtreated animals.In animals treated with PH plus NDMA,there was a marked increase in the numbers of BrdUlabelled cells compared with animals treated with NDMA alone from 5 hours to 1 week after injection of NDMA. BraUlabelled cells mainly gathered in the vicinity of portal tract.Conclusions: NDMA may stimulate the DNA synthesis of hepatocytes and hepatic sinusoidal wall cells,and PH plus NDMA possesses a synergetic effect for stimulating the DNA synthesis of hepatocytes and hepatic sinusoidal wall cells.
