摘 要:目的探讨甘氨脱氧胆酸钠(glycodeoxycholic acid,GDCA)对肝癌SMMC7721细胞端粒酶活性以及hTERTmRNA表达影响。方法采用PCR-ELISA法检测端粒酶活性;应用RT-PCR检测hTERTmRNA的表达。结果GDCA作用肝癌SMMC7721细胞后,端粒酶活性与hTERTmRNA表达均降低,GDCA200、400μmol/L处理组与对照组端粒酶活性(A450nmOD值)分别为(0.140±0.04)、(0.077±0.01)、(0.248±0.05),差异有统计学意义(P<0.05),抑制率为43.5%和69.0%。hTERTmRNA表达明显降低,相对表达率分别为1.33、0.59、1.89。结论甘氨脱氧胆酸钠抑制肝癌SMMC7721细胞端粒酶活性,且下调hTERTmRNA的表达。Objective To investigate the effect of telomerase activity and hTERT mRNA expression in SMMC7721 cells treated with GDCA in vitro.Methods Telomerase activity was examined by PCR-ELISA;The expression of hTERT mRNA was analyzed by RTPCR.Results Both of telomerase activity and hTERT mRNA expression were obviously deceased after the SMMC721 cell was treated by GDCA. The OD450nm of the groups of GDCA200,400μmol/L and control were ( 0.140 ± 0.04 ), ( 0.077 ±0.01 ), ( 0.248 ± 0.05 ) respectively( P 〈 0.05 ). The telomerase activity inhibition rate of the groups of GDCA200,400μmot/L was 43.5% and 69.0%. The relative expression rate of hTERT mRNA of groups of GDCA200,400μmol/L and control were 1.33,0.59,1.89 respectively. Conclusion GDCA inhibit the telomerase activity and downregulate the expression of hTERT mRNA in hepatocellular carcinoma cell SMMC7721.
