神经母细胞瘤中酪氨酸激酶受体和血管内皮生长因子基因表达相关性的研究

摘　　要：目的探讨神经母细胞瘤(NB)中酪氨酸激酶受体(TrkA,TrkB)和血管内皮生长因子(VEGF)的基因表达相关性及临床意义。方法采用逆转录聚合酶链反应(RT-PCR)法半定量检测51例NB肿瘤中TrkA,TrkB及VEGFmRNA的表达。结果TrkA的表达在低分期组明显大于高分期组(P<0.01),而VEGF的表达在低分期组明显小于高分期组(P<0.05),它们的表达水平呈明显负相关(r=-0.437,P<0.01),并与患者2年存活率之间具有明显的相关性(P<0.01);TrkB的表达在高分期组均明显大于低分期组(P<0.01),与VEGF的表达存在正的直线相关关系(r=0.342,P<0.05),也与患者2年存活率之间具有明显的相关性(P<0.01)。结论TrkA基因在预后较好的NB瘤中表达量高,TrkB与VEGF基因在预后差的NB瘤中表达量高;TrkA与VEGF基因表达成负相关,TrkB与VEGF基因表达成正相关。TrkA,TrkB与VEGF这3种基因的表达及其相关性对NB肿瘤的分期和预后评估具有重要的临床意义。Objective To study the correlativity and clinical significance of gene expression of tyrosine-kinase receptors （ TrkA, TrkB） and vascular endothelial growth factor （VEGF） in human neumblastomas. Methods Expression of TtkA, TrkB and VEGF mRNA was senti-quantitatively detected by reverse transcription-polymerase chain reaction（RT-PCR） in 51 cases of neuroblastomas. Results The expression of TrkA was significantly higher in lower-stage group than in higher-stage group （P 〈 0.05 ）, while the expression of VEGF was significantly lower in lower-stage group than in higher-stage group （P 〈 0.05 ）.The expression of TrkA was correlated negatively with the expression of VEGF （P 〈 0.01 ）,and had remarkably correlativity with 2-year cumulative-survival rate （P 〈 0.01 ）. The expression of TrkB was significantly higher in higher-stage group than in lower-stage group （ P 〈 0.05 ） and was positively correlated with VEGF expression（ r = 0.342,P 〈 0.05）,and their expressions also had remarkably correlativity with 2-year cumulative-survival rate （P 〈 0.01）. Conclusion TrkA is expressed high in neumblastomas with good prognosis; however TrkB and VEGF are expressed high in neuroblastomas with poor prognosis. The expression of TrkA is negatively correlated with the expression of VEGF, while the expression of TrkB is positively correlated with the expression of VEGF. These three genes have important clinical significance in tumor stage and outcome for patients with neumblastomas.
