人脑胶质瘤中Apaf-1基因启动子区甲基化的研究

摘  要:目的探讨凋亡蛋白酶活化因子1(Apaf-1)基因启动子区甲基化在脑胶质瘤中的表达及发生中的作用。方法应用半定量RT-PCR和甲基化特异性PCR分析方法分析37例脑胶质瘤及9例瘤旁非肿瘤脑组织中Apaf-1基因表达及启动子区甲基化情况。结果37例胶质瘤中23例Apaf-1mRNA表达下调,而瘤旁脑组织未发现Apaf-1mRNA表达明显下调或者上调;在Apaf-1基因表达明显下调的23例胶质瘤中15例出现甲基化,表达水平无明显下调的14例胶质瘤中仅4例出现甲基化,而瘤旁非肿瘤的9例脑组织中未检测到Apaf-1基因启动子甲基化。结论Apaf-1基因与脑胶质瘤的发生、发展相关,启动子区甲基化是该基因失活的重要机制。Objcetive To investigate the effect and promoter methylation of Apaf-1 gene in progression of gliomas. Methods The expression and promoter methylation of Apaf-1 gene were detected by semi-quantitative RT-PCR and Methylation specific PCR in 37 specimens of brain gliomas and 9 specimens of brain tissues near tumors. Results The expression of 23 specimens of glomas ( 23/37 ) was lower than that of the brain tissues near tumor; methylation specific PCR displayed that 15 specimens were methylated in promoter regions in 23 specimens gliomas,whose expression of Apaf-1 mRNA was down regulated. By contrast,only 4 of 14 gliomas without changes in Apaf-1 mRNA levels were methylated. Conclusion Apaf-1 plays a role in progression of gliomas. The Apaf-1 DNA methylation in promoter region might contribute to the decreased transcription of Apaf-1 in gliomas .
