摘 要:目的从行为学及磁共振成像(MRI)表现上评价6-羟基多巴胺(6-OHDA)单侧毁损大鼠黑质致密部建立偏侧帕金森病(PD)模型的方法。方法70只SD大鼠随机分为3组:A组,30只,15μg/3μl黑质致密部(SNc)单点注射;B组,30只,30μg/6μlSNc单点注射;C组10只6μl含0.2%抗坏血酸的生理盐水SNc单点注射。术后不同时期观察阿扑吗啡(APO)腹腔注射诱导的大鼠旋转行为。6周后各组随机选取部分成模与非成模大鼠在高场强MRI下活体检测,观察大鼠颅内毁损情况。结果A、B组大鼠6-OHDA毁损黑质后,大部分成模大鼠在第3天可以检测到稳定的旋转(>210r/30min),其余成模大鼠在第14天可以检测到稳定的旋转。两组成模率相仿,成模率高达76.4%。第6周,MRI显示成模大鼠穿刺针道恰位于黑质部,而非成模大鼠穿刺针道偏离黑质部。结论应用6-OHDA单侧毁损大鼠黑质致密部可以高效、简便、稳定地制作PD大鼠模型;MRI检测可以客观评价其成模质量。Objective To study the magnetic resonance imaging (MRI)analysis of rat model of Parkinson's disease (PD) induced by unilateral substantial nigra pars compacta(SNc)lesion with 6- hydroxydopamine(6-OHDA). Methods Seventy SD rats were randomly divided into three groups. 6-()HDA 15 μg/3μl (group A, 30 rats) or 30 μg/3 μl(group B, 30 rats) was injected in unilateral SNc. Ten rats in group C was injected 6μl normal saline with 0. 2% ascorbic acid in unilateral SNc instead of 6-OHDA as in the controls. The rolling behaviour of rats induced by apomorphinc(APO) was observed in different stages after operated. MRI analysis was performed on the randomly selected rats from each group, which were established PD models and not established PD models, for observing in vivo the changes of the SNc lesion six weeks later. Results After injection of 6-OHDA in two groups, the stable rolling behaviour(〉210 r/30 min) was seen in the established PD models mostly on the 3rd day and partly on the 14th day. The success rates of PD model establishment of two groups were similar with the highest rate of 76.4%. On the 6th week , MRI scaning showed that transfixion pin hole located in SNc in the PD model rats,which was not in the non-PD model rats. Conclusion Unilateral SNc lesion with 6-OHDA can establish the PD rats model with the advantages of high efficiency, convenience and stability. MRI scaning can be used objectively to evaluate the quality of PD rats models.
