摘  要:目的:研究电刺激小脑顶核(Fastigial nucleuse lectrostimulation,FNS)对阿尔茨海默病(Alzheimer’s diseases,AD)模型大鼠学习记忆能力及海马神经元凋亡的影响。方法:40只SD大鼠随机分成四组:Aβ1-40微量注射组(AD组)、假手术组(SC组)、Aβ1-40微量注射组+电刺激小脑顶核组(FNS组)和Aβ1-40微量注射组+电刺激齿状核组(DNS组),采用Morris水迷宫检测电刺激小脑顶核对AD大鼠认知功能的的影响;通过尼氏染色观察电刺激小脑顶核对AD大鼠海马神经元形态的影响;采用TUNEL凋亡染色法观察电刺激小脑顶核对AD大鼠海马神经元凋亡的影响。结果:与SC组比较,AD组、FNS组和DNS组每天隐匿平台逃避潜伏期明显增长(P<0.05)、海马CA1区TUNEL阳性细胞数明显增多(P<0.05);FNS组与AD组比较,空间学习记忆能力明显改善(P<0.05),海马CA1区TUNEL阳性细胞数分别为(22.55±2.74、36.90±4.07)两组比较差异显著(P<0.05),而DNS组无此作用。结论:FNS可改善Aβ1-40诱导的AD大鼠空间学习记忆障碍,可能与抑制海马细胞凋亡有关。Objective: To study the effects of the fastigial nucleus electrostimulation on the ability of learning and memory and the Apoptosis of the neurons in the hippocampal district in ratymodel of Alzheimer' s Disease induced by β -amyloid (Aβ ). Methods: Forty rats were randomly divided into:A β 1-40 micro-injection groups ( AD ), sham-operation roup ( SC ), A β1-40 micro-injection and fastigial nucleus stimulation group( FNS ), a Aβ1-40 microinjection and corpus dentatum stimulation group( DNS ). The effect of fastigial nucleus electrostimulation on learning and memory retention disorder in the model rats was studied by the test of Morris water maze;the brain protection of fastigial nucleus electrostimulation in model rats was observed through pathomorphology changes ; the Apoptosis of the neurons in the hippocampal CA1 district was detected by Terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling method. Results: The escape latency of the place navigation in the AD grou, FNS group and DNS group were singnificiantly longer than that in the SC group (P〈0.05).The number of apoptotic neurons in the hippocampal CA1 district of the AD group was significantly different from that in the sham injection group(P〈0.05 ). The AD rats received FNS showed significiant shorter escape latency than AD rats(P〈0.05 ). The number of apoptotic neurons in the hippocampal CA 1 district of the FNS group(22.55 ± 2.74) was significantly different from that of the AD group (36.90 ±4.07 ) (P〈 0.05 ). Conclusions: FNS can improve the spatial learning and memory dysfunction of AD rat, implying that FNS has the function to restraining the apoptosis of the hippocampal neurons.
