摘 要:目的研究拟老年痴呆症(alzheimer’s disease,AD)模型大鼠学习记忆功能的改变及丹参酮的治疗作用。方法(1)实验动物随机分成4组。AD模型组:采用D-半乳糖腹腔注射和大鼠双侧海马齿状回背侧微量注射凝聚态β-淀粉样肽1-40(amyloid beta-peptide,Aβ1-40)的复合造模方法,拟建立AD样学习记忆障碍动物模型;丹参酮治疗组:AD大鼠造模24 h后给予丹参酮[50 mg/(kg.d)],溶于5 mL玉米油中灌胃14 d;假造模对照组:与AD大鼠同法注射等量生理盐水;正常对照组:大鼠不做任何处理。(2)采用水迷宫行为学实验检测各组大鼠的学习记忆功能。结果(1)AD模型大鼠发生了空间学习记忆功能障碍,与两对照组比较有显著性差异(P<0.01)。(2)丹参酮治疗组治疗后较AD模型组潜伏期缩短、误入盲端次数也明显减少。结论(1)D-半乳糖-Aβ复合造模致AD样学习记忆功能障碍的动物模型具有较好的仿真AD的特点。水迷宫实验能较真实地反应动物学习记忆功能。(2)丹参酮对AD样大鼠的学习记忆功能障碍具有一定的改善作用。Objective To study the learning and memory Impairment of alzheimer's-like disease model rats and Curative effectiveness of tanshinone. Methods (1) The experimental animals were randomly devided into four groups:In AD model group: by using intraperitoneal injection of D-galactose in combination with microinjection of 10 μg incubated amyloid beta-peptide1-40 (Aβ1-40) into the dorsal blade of the dentale gyrus in the hippocampus of rat, AD-like model was established, in tanshinone therapeutic group: after establishment of rat AD models 24 h , tanshinone [50 mg/(kg · d)]dissolved in 5 mL maize oil was given for 14 days by gastric perfusion. In false model group: the rats were treated with the same method as AD rats but receiving injection of the same physiological saline. In normal control group:the rats were not treated. (2) Learning and memory function of various group rats were assessed by using a water-maze test. Results (1) Learning and memory function of AD model rats had impaired significantly as compared with two control groups. (2) After tanshinone therapeutic group was treated with transhinone, escape latency was obviously shortened and frequency of escape failure was decreased as compared with AD model group. Conclusion (1)AD-like rat models of learning and memory Impairment induced by D-galactose combination with Aβ1-40 have preferably characters of aging-dementia. A watermaze test could assess accurately learning and memory function of rats. (2)Tanshinone improves respectively to AD-like rats on learning and memory Impairment.
