摘  要:目的探讨不同CO2气腹压力对人宫颈癌CaSki细胞体外生长的影响。方法建立CO2气腹体外模型,以不同压力(0、7、14 mm Hg)的CO2气腹对人宫颈癌CaSki细胞处理4 h,设未处理组为对照组。采用MTT法、流式细胞术及电镜比较各组CaSki细胞的生长情况。结果7 mm Hg组第1-5天吸光度值明显高于0 mm Hg组、14 mm Hg组、对照组,差异有统计学意义(P<0.05);7 mm Hg组G2/M期的人宫颈癌CaSki细胞数值明显高于0 mm Hg组、14 mm Hg组、对照组,差异有统计学意义(P<0.05);14 mm Hg组S期人宫颈癌CaSki细胞数值明显高于0 mm Hg组、7 mm Hg组、对照组,差异有统计学意义(P<0.05);人宫颈癌CaSki细胞的增殖指数随着CO2气腹压力的升高而增高;电镜下观察结果显示,7 mm Hg组、14 mm Hg组均可见明显异常的核分裂像。结论CO2气腹在一定的压力状态下表现出较强的促细胞生长效应。Objective To evaluate the influence of CO2 pneumoperitoneum with different pressures on the growth of human cervical carcinoma cells in vitro. Methods The CO2 pneumoperitoneum model was established in vitro. After CaSki cells were exposed to CO2 pneumoperitoneum at 0 mm Hg,7 mm Hg or 14 mm Hg for 4 h with the untreated group as control,the cell growth was determined by MTT assay, flow cytometry and electron microscopy. Results The absorbance values of CaSki cells from the 1st clay to the 5th day of 7 mm Hg group were significantly higher than those of other groups(0 mm Hg group, 14 mm Hg group,control group) with a statistical significances(P〈0.05). The numerical values of CaSki cells in G2/M phases of 7 mm Hg group were significantly higher than those of other groups(0 mm Hg group, 14 mm Hg group,control group) ,and the differences had the statistical significances(P〈0.05). The numerical value of CaSki cells in S phase of 14 ram Hg group was significantly higher than those of other groups (0 mm Hg group,7 mm Hg group,control group), and the differences had the statistical significances(P〈0.05). The proliferation index increased with the pressure of CO2 pneumoperitoneum being raised. The result observed by electron microscopy showed that there were obvious abnormal karyokinesis of CaSki cells in both 7 mm Hg group and 14 mm Hg group. Conclusion Carbon dioxide pneumoperitoneum under certain pressures could increase the growth of human cervical carcinoma cells in vitro.
