摘 要:目的:探讨虎栀合剂(Hu Zhi Mixture,HZM)防治α-异硫氰酸萘酯(α-maphthy1 Isothiocyanate,ANIT)致大鼠急性肝损伤的效应及机制。方法:将大鼠随机分为4组:正常对照组、造模对照组、虎栀合剂治疗组、泼尼松对照组。检测大鼠在造模48h后,血清中肝功能、肿瘤坏死因子-α(TNF-α)、肝组织中的丙二醛(MDA)、超氧化物歧化酶(SOD)的含量,光镜下观察肝脏组织学改变。结果:HZM能明显改善造模引起的肝组织学及肝功能变化,降低TNF-α、MDA水平,而SOD含量显著增高。泼尼松对照组能部分改变各项指标。结论:HZM防治ANIT致大鼠急性肝损伤切实有效。其作用机制可能与其拮抗作用,减少TNF-α、MDA的产生,提高SOD活性有关。Objective :To study the therapeutic effects and mechanism of Hu Zhi Mixture (HZM) on rat model of acute liver injure induced by α-Naphthyl iso-thiocyanate (ANIT). Methods: The rats were randomly divided into four groups: normal group, ANIT group, HZM group and Prednison group. The rat model was established by ANIT. 48hours later, we determined the levels of tumor factor-α ( TNF-α ), malondialdehyde ( MDA), superoxide disnutase (SOD), examined liver functions and liver tissues under the microscope. Results:Compared with ANIT group, the levels of TNF-α ,MDA were markedly lowered in HZM group while the level of SOD markedly increased. Besides, HZM can significantly offset the changes in liver tissue and function induced by ANIT. Prednisone can improve the above values to a certain degree. Conclusion: HZM has good preventive and therapeutic effect against acute liver injure of rats which may be linked with inhibition of TNF-α and MDA, activation of SOD activity.
