摘 要:目的:观察42℃温热对人卵巢癌SKOV-3细胞热耐受和热耐受预测因子(HSP70)表达的影响。方法:(1)四氮唑蓝(MTT)快速比色法测细胞增殖率;(2)免疫细胞化学法测定HSP70的表达。结果:(1)42℃首次加热对人卵巢癌细胞SKOV-3有抑制和杀伤作用,短时间内(48h内)重复加热细胞,细胞会出现生长加快,以间隔8h重复加热组细胞生长加快尤为明显;(2)42℃加热SKOV-3细胞100min后48h内测HSP70表达,有不同程度的增加,以加热后8h细胞内HSP70的表达最强,有时间依赖性。结论:42℃加热100min可诱导人卵巢癌SKOV-3细胞内HSP70高表达,使细胞产生热耐受性,其出现与消退有时间依赖性。Objective:To investigate the effects of 42℃ hyperthermia on the thermotolerance and the expression of HSP70 in the human ovarian cancer cell line SKOV-3. Methods: (1)The proliferation rate of cells was detected by methyltetrazolium (MTF) assay. (2)The expression of HSP70 was detected by immunocytoehemical assay. Results: (1)Human ovarian cancer cell line SKOV-3 was inhibited and killed after 42℃ hyperthermia firstly. But the proliferation of the cell reheated in 48 hours was quickened, especially when reheated at 8h after first hypertherimia after 8 hours it increased obviously. ( 2 ) The expression of HSP70 in SKOV-3 cells increased differently at different time during 48 hours after 42℃ hyperthermia for 100 mins,with the maximal expression after 8 hours. The expression exhibited a time-dependent manner. Conclusion:42℃ hyperthermia for 100 mins can induce high expression of HSP70 and thermotolerance of the human ovarian cancer cell line SKOV-3. The thermotolerance appears and regresses in a time-dependent manner.
