摘 要:①目的观察大鼠肢体缺血再灌注(LIR)后骨骼肌损伤及缺血预适应(IPC)保护作用,并探讨IPC的可能保护机制。②方法实验用雄性Wistar大鼠24只,随机分为对照(Control)组、缺血再灌注(IR)组和缺血预适应(IPC+IR)组,每组8只。分别测定血浆乳酸脱氢酶(LDH)、肌酸激酶(CK)、血栓素B2(TXB2)、6-酮-前列腺素(6-Keto-PGF1α)的含量及TXB2/6-Keto-PGF1α比值的变化;测定骨骼肌组织中ROS、MDA、谷胱甘肽过氧化物酶(GSH-PX)、还原型谷胱甘肽(GSH)、Ca2+、Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶的变化及DNA双链百分率(Rat io of DNA Chain)的变化。③结果IPC+IR组血浆LDH、CK、TXB2/6-Keto-PGF1α比值及骨骼肌组织中ROS、MDA、Ca2+明显低于IR组,而骨骼肌组织中GSH-PX、GSH、Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶及DNA双链百分率明显高于IR组。④结论IPC减轻了LIR后骨骼肌的损伤。抑制自由基生成和提高自由基清除酶的活性,降低钙超载及改善TXA2/PGI2的平衡关系可能是IPC抗损伤的机制。Objective To investigate the injury of skeletal muscle following limbs ischemia - reperfusion (LIR) and the protective effects of ischemic preconditioning (IPC) in rats. Methods Wistar rats ( 24 cases) were randomly divided into control group, IR group, IPC +/R group ( each group 8 rats ). The contents of lactate dchydrogenase ( LDH), creatine phosphate kinase (CK) ,Thromboxane B2 ( TXB2 ), 6 - keto - prostaglandin F1α ( 6 - Keto - PGF1α ) and TXBJ6 - Keto - PGFI= in plasma were measured. The changes of ROS, MDA, glutathione peroxidase ( GSH - PX), glutathione ( GSH), Ca^2 + , Na ^+ - K^+ - ATPase, Ca^2 + - Mg^2+ - ATPase and the ratio of DNA double chain in skeletal muscle were measured. Results The values of LDH, CK,TXB2/ 6 - Keto - PGF1α of plasma and MDA, ROS, Ca^2+ of skeletal muscle in IPC + IR group were significantly lower than those in IR group, while Compared with IR group, the values of GSH - PX, GSH, Na^+ - K^+ - ATPase, Ca^2 +- Mg^2+- ATPase and the ratio of DNA double chain in IPC + IR group were obviously higher than those in IR group. Conclusion The injury of skeletal muscle following LIR was alleviated by IPC, its protective mechanism could be the calcium overload reduced and the equilibrium of TXBJ6 - Keto -PGF1α regulated to have effects on skeletal muscle following the LIR injury in rats.
