摘  要:目的观察5-烯丙基-7-二氟亚甲基白杨素(ADFMChR)对人卵巢癌CoC1裸鼠移植瘤生长的抑制及促凋亡作用。方法建立人卵巢癌CoC1裸鼠皮下移植瘤模型,随机分成5组:生理盐水组、顺铂组(2mg/kg)、低剂量ADFMChR组(6mg/kg)、中剂量ADFMChR组(18mg/kg)和高剂量ADFMChR组(54mg/kg),每组4只。观察各组裸鼠移植瘤生长情况、裸鼠体重的变化;PI染色流式细胞术(FCM)测定移植瘤细胞凋亡率。结果ADFMChR对移植瘤的生长有明显抑制作用,低剂量组、中剂量组和高剂量组的瘤重抑制率分别达62.76%、91.76%和77.55%。PI流式细胞术结果显示ADFMChR诱导移植瘤细胞凋亡,呈剂量依赖性。结论ADFMChR具有抑制人卵巢癌裸鼠移植瘤生长的作用并能诱导移植瘤细胞凋亡。[Objective] To investigate the inhibitory effects of 5-allyl-7-gen--difluoromethylene- chrysin (ADFMChR) on the growth of human ovarian carcinoma xenografts and its promotion on apoptosis in nude mice. [Methods] Model of human ovarian cancer transplanted subcutaneously in nude mice was established, and divided into 5 groups: control group, DDP (2 mg/kg) group, low dose ADFMChR (6 mg/kg) group, media dose ADFMChR (18 mg/kg) group and high dose ADFMChR (54 mg/kg) group. Tumor volume and weight were measured. The rate of apoptotie cells of human ovarian carcinoma xenografts was analyzed by PI stained flow eytometry (FCM). [Results] ADFMChR could signifieandy suppress tumor growth of human ovarian carcinoma xenografts in mouse models. After treatment with ADFMChR (6, 18 and 54 mg/kg), the inhibitory rates of the weight of human ovarian carcinoma xenografts were 42.86%, 62.76% and 77.55% respectively. [Conclusion] ADFMChR suppresses implanted tumor of human ovarian carcinoma ceils in nude mice through inducing apoptosis.
