Fas相关磷酸酯酶-1在肝癌中的表达及其临床意义

摘　　要：目的观察肝癌中Fas相关磷酸酯酶（FAP-1）的表达，探讨FAP-1表达与增殖和凋亡的关系，并分析其意义。方法采用免疫组织化学SP法检测60例肝癌及15例癌旁肝硬化组织标本中FAP-1蛋白的表达水平，设置10例正常肝组织为对照组。结果FAP-1蛋白的阳性表达主要定位在细胞质内，少数也可见胞膜着色。FAP-1蛋白在正常肝组织中未见表达，癌旁肝硬化组织中的表达率为6．7％，而在恶性肿瘤中的阳性表达率分别为78．3％，与前两者比较差异有统计学意义（P〈0．01）。FAP-1的表达率随肿瘤的转移临床分期晚和肿瘤分化程度低而增加，并与分化程度和转移性显著相关（P〈0．05），而与年龄和肿瘤大小无明显相关（P〉0．05）。结论FAP-1在肝癌组织中有广泛表达，且在临床有转移分期晚和病理分级低的组织中表达增高。Objective To investigate the roles of Fas-associated phosphatase-1 （ FAP-1 ） in the pathogenesis of hepatocellular carcinoma （HCC）. Methods Immunohistochemical SP method was employed to detect the expression of FAP-1 in para-cancerous tissue （ 15 cases ）, HCC （ 60 cases ） and the normal controls （ 10 cases）. In addition, clinicopathological data of them were analyzed. Results There was no FAP-1 expression in normal controls and low positive expression rate in para-cancerous tissue （6.7%） ,but high positive expression rate （78.3%） was found in HCC （P 〈 0.01 ）. Moreover, the expression of FAP-1 was markedly associated with metastasis and tumor pathological grade （ P 〈 0.05 ）, but not with age and tumor size. Conclusion FAP-1 is widely overexpressed in HCC, and its expression is associated with metastasis and pathological grade.
