摘 要:目的以子宫鳞癌SiHa细胞为靶细胞,分析甘氨双唑钠是否可影响宫颈鳞癌SiHa细胞株对顺铂的敏感性。方法不同浓度顺铂、甘氨双唑钠及两药联合作用于SiHa细胞,通过形态学观察、四甲基偶氮唑盐细胞活力测定等方法检测对SiHa细胞敏感性的影响。结果不同浓度的顺铂对宫颈癌细胞的生长均有抑制作用(P〈0.05),而不同浓度甘氨双唑钠对SiHa细胞影响不显著(P〉0.05)。顺铂(5μg/mL)与不同浓度CMNa合用可明显增强顺铂对SiHa细胞生长的抑制作用,且随CMNa浓度的增加,组间抑制作用越明显(P〈0.01)。结论联合应用甘氨双唑钠可提高顺铂对肿瘤细胞的敏感性,为临床制定宫颈癌化疗方案提供理论依据。Objective To determine whether CMNa (a novel tumor chemosensitizer) can influence chemosensitivity of target cells - cervical squamous carcinoma cell line (SiHa cells) to cisplatin. Methods SiHa cells were subjected to cisplatin (PDD) and CMNa in different concentrations for 24 hours, and morphology of the SiHa cells were observed. MTT colorimetry was used to determine influences of two drugs on SiHa cells. Results Different concentrations of cisplatin presented an inhibitory role on proliferation of the SiHa cells ( P 〈 0.05 ). While influences of different concentrations of CMNa on the SiHa cells were not significant ( P 〉 0.05 ). The combination of CMNa and cisplatin can enhance the inhibitory role on SiHa cells greatly. And as increase of CMNa concentration, the inhibitory effect was more significant as compared with other groups (P 〈0.01 ). Conclusion CMNa can enhance the sensitivity of tumor cells to PDD, which provides a theoretical basis for formulating chemotherapy plan for patients with cervical carcinoma.
