摘 要:目的:研究血管内皮细胞生长因子(VEGF)及其受体2(VEGFR-2)在兔正畸牙周组织中的表达及其变化的相关性。方法:35只大耳白兔随机分成7组:正常组及实验1、3、5、7、14、21d组,每组5只。实验组动物于双侧上颌第一磨牙至切牙间拴结镍钛螺簧,0.08N力拉上颌第一磨牙向近中移动。对实验标本行HE染色、VEGF、VEGFR-2免疫组化染色,进行组织学观察和计算机图像分析,对VEGF、VEGFR-2表达的灰度积分进行统计学处理。结果:正常牙周组织中VEGF及VEGFR-2有少量表达。实验1d组~7d组压力区VEGF及VEGFR-2的表达明显高于正常牙周组织,有显著性差异,峰值均出现在5d组;实验1d组张力区VEGF及VEGFR-2表达与正常组相比无明显差别;实验3d组~14d组张力区VEGF及VEGFR-2表达均高于正常牙周组织,VEGF的表达峰值出现在14d组,VEGFR-2的表达在7d组出现峰值。无论在压力区还是在张力区,VEGF与VEGFR-2的表达均存在正相关关系。结论:在兔正畸牙齿移动过程中牙周组织VEGF及VEGFR-2的表达增强,VEGF与VEGFR-2的表达呈正相关;VEGF及VEGFR-2参与了正畸牙齿移动过程中牙周组织的改建过程。Objective: To investigate the expressions of vascular endothelial growth factor(VEGF) and VEGF receptor 2(VEGFR-2) in the periodontal tissues during experimental teeth movement in rabbits, and analyze the role of VEGF and VEGFR-2 in orthodontic periodontal vascularization and bone remodeling. Methods :Thirty-five rabbits were randomly divided into 7 groups: Normal group and experimental group of 1, 3, 5, 7, 14, 21 days. Bilateral maxillary first molars and incisors were ligated with orthodontic wire and a coil spring was inserted between them, and then exerted by a force of approximately 0.08 N. lmmunohistochemical staining was used to detect the expressions of VEGF and VEGFR-2. The staining intensity was evaluated by microphotometry and analyzed by the statistical software. Results:The VEGF and VEGFR-2 was found in PDL of rabbit molars in normal group. VEGF was expressed in newborn vascular endothelial cells, no expression was found in mature vascular endothelial cells. Compared to normal group, the expressions of VEGF and VEGFR-2 in experiment group significantly increased at 1 day and reached peak value at 5 days in the pressure region, but in the tension region, the expressions of VEGF and VEGFR-2 increased at 3 days and VEGF reached peak value at 14 days while VEGFR- 2 reached peak value at 7 days. The expressions of VEGF and VEGFR-2 have positive correlation both in the pressure region and in the tension region. Conclusion: The VEGF and VEGFR-2 expressions in periodontal tissues are promoted by mechanical stimuli during experimental tooth movement in rabbits. VEGF expression is correlated positively with expression of VEGFR-2. VEGF and VEGFR-2 participate in the tissue remodeling during experimental tooth movement.
