摘 要:目的观察顺铂联合足叶乙甙(PE方案)对卵巢癌细胞增殖和细胞周期的影响,并探讨其作用机制。方法体外培养的卵巢癌SKOV3细胞,加入不同浓度的顺铂和足叶乙甙,作用24 h。MTT法检测细胞抑制率;流式细胞仪测定细胞周期,免疫细胞化学法检测细胞的Cyclin B1、p34cdc2和survivin。结果足叶乙甙和不同浓度的顺铂联合应用,可明显抑制SKOV3细胞的增殖,S期和G2-M期细胞增加,G0-G1期细胞减少,细胞Cyclin B1、p34cdc2和survivin表达下调。结论顺铂联合足叶乙甙对SKOV3细胞具有协同抑制作用,阻滞细胞周期,其机制可能与细胞Cyclin B1p、34cdc2和survivin的表达下调有关。Objective To observe the effect of cisplatin combined with etoposide on the proliferation and cell cycle of human ovarian carcinoma cell line, and investigate the mechanisms. Methods Ovarian cancer cell line SKOV3 was interfered with cisplatin and etoposide in different concentration for 24 h.The proliferation inhibition rate was measured by MTT, the cell cycles were analyzed by the flow cytometry, the C yclin Bi , p34^cdc2 and survivin were determined by immunnhistochemistry method. Results Etoposide combined with different concentration cisplatin inhibited the proliferation of SKOV3 cell, S and G2 - M phase cell percentage increased, while Go - G1 phase decreased, the expression of Cyclin B1, p34^cdc2 and survivin were decreased. Conclusions Etoposide combined with cisplatin shows significant synergistic inhibition on SKOV3 cell line, the mechanism may be related with the degressive expression of Cyclin B1 , p34^cdc2 and survivin.
