胆管癌组织、细胞中CXCR3 mRNA及其蛋白的表达变化

摘  要:目的检测趋化因子受体3(CXCR3)在胆管癌中的表达,并探讨其意义。方法采用逆转录聚合酶链反应(RT-PCR)、免疫细胞化学和Western-Blot方法检测胆管癌细胞系QBC939和ICC-9810中的CXCR3 mRNA及其蛋白;采用免疫组化SP法检测人胆管癌组织和癌旁组织中的CXCR3蛋白,并分析探讨其与胆管癌分化程度及淋巴结转移的关系。结果QBC939和ICC-9810中CXCR3 mRNA和蛋白表达均呈阳性;胆管癌组织中CXCR3蛋白阳性表达率为80%,癌旁组织中没有检测到CXCR3蛋白,CXCR3蛋白与胆管肿瘤分化程度和淋巴转移有关(P<0.05)。结论CXCR3在胆管肿瘤细胞系中呈阳性表达,CXCR3和胆管肿瘤的分化及侵袭转移有一定关系。Objective To detect the expression of CXCR3 in human cancer of bile duct tissue and cell and investigate the mechanism. Methods The expression of CXCR3 mRNA and protein in cholangiocacinoma cell line QBC939 and ICC-9810 was detected by RT-PCR,Westem-blot assays,ICC. The CXCR3 protein expression was detected by immunohisto-chemistry in human cancer of bile duct and matched normal tissue. The relationship of CXCR3 with the stage of the pathology and metastasis of the lymph nodes was explored by analyzing the case and the result of the immunohistochemistry human cancer of bile duct. Results CXCR3 mRNA and protein was postive expressed in cholangiocacinoma cell line QBC939 and ICC-9810. The positive rate of CXCR3 in human cancer of bile duct was 80.0% (24/30), whereas the protein in matched normal tissue was not detected. CXCR3 was correlated with tumor differentiation san lymph node metastasis (P 〈 0.05). Conclusions CXCR3 plays an important role in genesis and development of human cancer of biliary tract. There is corre-lation between the protein expression and the malignancy degree of pancreatic carcinoma.
