摘  要:目的观察苦参碱(Ma)诱导结肠癌SW1116细胞凋亡的作用,并探讨其机制。方法采用不同浓度的Ma处理SW1116细胞48h,并设正常对照组,MTT法检测细胞增殖抑制率,流式细胞仪检测细胞凋亡率,半定量RT-PCR检测Fas和Bax mRNA表达水平。结果与正常对照组相比,不同浓度Ma处理的SW1116细胞抑制率、胞凋亡率显著升高(P均(0.01),Fas和Bax mRNA表达上调(P<0.05,P<0.01)。结论Ma可诱导SW1116细胞凋亡,其作用机制可能与上调Fas和Bax mRNA表达有关。Objective To observe the effects of Matrine(Ma) on induction of apoptosis in colon cancer SW1116 cells and explore the mechanism. Methods Human colon cancer SW1116 cells were treated with different dose of Ma for 48h. Besides, control group was established. The proliferation inhibitory rate was evaluated by MTT assay. Apoptotie rate was determined by flow cytometry. The expression of Fas and Bax mRNA were detected by semi-quantitive RT-PCR. Results Compared with control group, the inhibitory rate, apoptotic rate increased obviously(P〈0.01), the expression of Fas and Bax mRNA up-regulated in different dose Ma groups. Conclusion Ma can induce the apoptosis of SW1116 cells, and its molecular mechanism may be associated with up-regulating the expression of Fas and Bax mRNA.
