摘  要:研究了十八烷酰基乙醛亚硫酸钠系列化合物中双键数目对兔红细胞溶血,膜脂流动性及膜蛋白构象的影响.结果表明:1.81×10-4mol/L的HOU-C18:0和2.42×10-4mol/L的HOU-C18:1即能引起细胞溶血,而HOU-C18:2在测试的浓度范围内未引起溶血.随着双键个数的增加,膜脂流动性迅速增加.对于HOU-C18:0,膜脂流动性变化的转折点与开始溶血的浓度是一致的.表明溶血与膜脂结构的变化有关.膜蛋白荧光结果表明,HOU-C18:n影响了膜蛋白的构象.膜蛋白与HOU-C18:n的结合数随着双键个数的增加而减少.双键数目增加,其疏水活性下降.离体实验表明HOU-C18:n是通过与膜脂和膜蛋白的疏水相互作用来发挥其药理功能的.The effects of double-bond number in octadecanoyl acetal sodium sulfite (HOU-C18,n ; n= 0, 1 and 2) on the hemolysis of erythrocytes, the fluidity of membrane and the conformation of membrane protein were investigated. The results showed that 1.81 × 10^-4 mol/L of HOU C18:0 and 2.42 × 10^-4 mol/L of HOU-C18,1 could induce cell plasmatorrhexis. Fluidity of the membrane lipid increased quickly with the increase in double-bond number. For HOU-C18:0, the turning point of the fluorescence polarization (P) value coincided with the concentration at which the erythrocytes began to crack, thus indicating that HOU-C18,n changed the structure of lipid bilayer and caused the membrane to crack. The results of fluorescence experiments indicated that HOU-C18,n influenced the conformation of the membrane protein. The binding number of the membrane protein with HOU-C18,n decreased with the increase in double-bond number. The interaction of HOU-C18,n with membrane lipid and protein was related with their hydro- phobic activity. In vitro experiments suggested that modifying the structure of cell membrane by hydrophobic interaction would be one of the important factors for HOU-C18:n to exert its pharmaceutical effects.
