摘 要:本文研究了利用免疫化学方法鉴别小麦胚乳贮藏蛋白高分子量谷蛋白亚基1Dx5+1Dy10的可能性。结果表明:1Dx5+1Dy10和1Dx2+1Dy12亚基与单克隆抗体的反应不同,利用特异性单克隆抗体可以鉴别小麦胚乳贮藏蛋白中是否含有1Dx5+1Dy10亚基,鉴别的准确度受抗体种类、抗体浓度及包被蛋白浓度的影响;HMW-GS在F_1籽粒中表现为倾母遗传。免疫化学测定法具有快速、简单、准确、所需样品少等特点,可用于育种早代的辅助选择。The possibility of differentiating HMW-GS 1Dx5+1Dy10 with immunochemi-cal method was studied with the following results; The speciality of 1Dx5+1Dy10 and 1Dx2 +1Dy12 to monoclonal antibodies against 1Dy10 were different, 1Dx5+1Dy10 could be distinguished from 1Dx2 + 1Dy12 with specific monoclonal antibodies, and the differentiating of 1Dx5+1Dy10 from 1Dx2 + 1Dy12 was affected by different kinds of monoclonal antibodies and thedillution of antibodies and coating proteins. The inheritance of 1Dx5+1Dy10 shows manternaleffect. EIISA has the characteristics of fasting, simplicity and accuracy, which could be used toassist selection for high quality crosses in early generation of breeding program.
