摘 要:单核细胞增生李斯特菌(Listeria monocytogenes,LM)是一种重要的食源性致病菌,能引起人和动物的李斯特菌病,利用双向电泳通过裂解液组成、上样量、聚焦时间等相关技术的比较研究和条件优化,获得单核增生性李斯特菌菌体全蛋白双向电泳图谱,提取部分蛋白质点酶解,利用基质辅助激光解吸电离飞行时间质谱(matrix-assisted laser desorp-tion/ionization time of flight mass spectrometry,MALDI-TOF-MS)测定肽质量指纹图谱,Mascot软件查询Swiss-Prot数据库,最终鉴定出李斯特菌中特异性蛋白包括核糖体蛋白L10、S6,李斯特菌推测蛋白LMOf2365_2340,ArsC蛋白,推测蛋白lin1505、lin1144,翻译起始因子IF-1等,反映出李斯特菌的多种差异及生长特性。Listeria monocytogenes(LM) is a foodborne pathogen,which can result in meningitis,cardiomyopathy,sapraemia,stillbirth or premature delivery in both human being and animals.We have applied proteomic techniques in this work to globally analyze the protein expression profiles of Listeria monocytogenes(LM),aiming at elucidating the components of Listeria monocytogenes(LM) replication and the cellular function at stationary growth station.The protein mixtures of three subcellular fractions from Listeria monocytogenes(LM) were separated by 2-DE under pH 3-10 gradients.Ten of Expressed spots were identified by MALDI-TOF MS,followed by database searching including proteins associated with host cytoskeleton,intracellular traffic,oxidative and translation,proliferation,etc.Our study provides the global information of proteomic expression of Listeria monocytogenes(LM) cells and the clues for further understanding of the mechanism of Listeria monocytogenes(LM)s at stationary growth phase.
