热休克蛋白70、90基因在糖皮质激素抵抗型哮喘患者血单个核细胞中的表达

摘　　要：目的探讨热休克蛋白（ＨＳＰ）７０、９０α、９０β信使核糖核酸（ｍＲＮＡ）在糖皮质激素抵抗型（ＳＲ）哮喘发病机制中的作用。方法分激素抵抗型（ＳＲ）哮喘组９例，激素敏感型（ＳＳ）哮喘组１６例，正常对照组１０例，采用反转录－聚合酶链反应（ＲＴＰＣＲ）法测定研究对象外周血单个核细胞（ＰＢＭＣ）中的ＨＳＰ７０、９０α、９０βｍＲＮＡ的表达；利用淋巴细胞增殖抑制试验观察地塞米松（Ｄｅｘ）对研究对象淋巴细胞增殖的抑制程度。结果正常人的ＰＢＭＣ中不表达ＨＳＰ７０ｍＲＮＡ。ＳＲ哮喘患者中ＨＳＰ７０、９０α、９０βｍＲＮＡ的表达依次为２９５±１１２、２１７±０８９、２２２±０８３明显高于ＳＳ哮喘患者ＨＳＰ７０、９０α、９０βｍＲＮＡ的表达，依次为０２３±００９、１０７±０３９、０９４±０３２（Ｐ＜０．０１）。两者的ＨＳＰ９０α、９０βｍＲＮＡ表达均显著高于正常人依次为０４５±０１９、０３２±０１５（Ｐ＜０．０１）。ＳＳ、ＳＲ哮喘患者的ＨＳＰ７０、９０α、９０βｍＲＮＡ表达与Ｄｅｘ为１０７ｍｏｌ／Ｌ时的淋巴细胞增殖抑制程度之间的直线相关分析呈显著正相关（Ｐ＜０．０１）。结论ＳＲ哮喘患者的ＰＢ?Objective To investigate the role of heat shock protein(HSP)70、90α、90β mRNA on the pathogenesis of steroid resistant(SR) asthma. Method 9 SR asthmadics 16 steroid sensitive (SS) asthmatics and 10 normal healthy volunteers were studied. With reverse transcription polymerase chain reaction(RT PCR),the expression of hsp70、90α、90β mRNA were detected in peripheral blood mononuclear cell(PMBC) from normal volunteers and SR. steroid sensitive(SS) asthmatics. Inhibition of PHA induced lymphocyte proliferation by dexamethasones (Dex) was investigated using lymphocyte proliferation assay. Result There were no expression of hsp70mRNA in PMBC of normal volunteers; the levels of expression of hsp70,90α,90β mRNA in PBMC of SR asthmatics (70=2.95±1.12, 90α=2.17±0.89, 90β=2.22±0.83) were significantly higher than SS asthmatics (70=0.23±0.09, 90α=1.07±0.39, 90β=0.94±0.32) ( P <0.01) and normal volunteers (70=0, 90α=0.45±0.19, 90β=0.32±0.15) ( P <0.01); There were significantly positive correlation between the degree of inhibition by Dex(10 7 mol/L) of SR、SS asthmatics of proliferation of T lymphocytes and the expression of hsp70、90α、90β mRNA in PMBC( P <0.01). Conclusion There were expression of hsp70、90α、90β gene in PMBC from SR asthmatics ,but increased with worsening of inflammation and prolonging of the duration of disease. The more hsp 90 gene expression ,the less T lymphocytes were inhibited by Dex.
