摘 要:目的:探讨由唐传(厦门)生物科技有限公司从水稻,玉米秸秆中提取的L-阿拉伯糖在降低空腹血糖,提高糖耐量及控制体重方面的重要生物学功能,从而为其进一步的功能食品开发提供科学依据。方法:以20g~22g昆明小鼠为实验动物,在给予高糖高脂饲料喂养的同时,采用高、中、低三种剂量口服L-阿拉伯糖水溶液一个月,以单独喂养高糖高脂饲料喂养组为对照,实验期间检测小鼠体重变化,食量。4周结束后进行小鼠空腹血糖及蔗糖糖耐量试验。结果:显示高剂量(250mg/只)和中剂量组(150mg/只)对小鼠空腹血糖有明显降低作用(P<0.05),而低剂量组(30mg/只)作用不明显,糖耐量试验证明三个剂量组的L-阿拉伯糖都具有明显增强小鼠糖耐量作用,与对照组有极显著差异(P<0.01),实验过程中未发现实验组与对照组小鼠食量有明显差异。解剖结果发现非用药组肝组织外观颜色淡黄色,而喂养L-阿拉伯糖组肝脏颜色为正常的深红色,肝指数变大,病理切片发现,实验对照组肝组织结构发生改变,肝细胞脂肪积累,PAS染色发现,肝脏切片染成深红色,而喂养L-阿拉伯糖组PAS染色成浅红色。结论:L-阿拉伯糖能够明显降低喂养高糖高脂饲料小鼠的空腹血糖,增强糖耐量,并且能够有效减缓体重增长趋势。组织切片和PAS染色证明L-阿拉伯糖可以有效抑制外源性糖在肝脏细胞中的积累及相应造成的脂肪堆积,进而对糖在体内转化成脂肪从而造成脂肪在体内的堆积也有重要的抑制作用。Objective:This study is to examine the biological activity of L-Arabinose and provide experimental basis to further the development of functional foods.Methods: 100 four-week male Kunming mice were randomly divided into 4 groups,A,B,C,Dgroup,20 mice one group,D group fed with high fat and high sucrose diet only as control group,A,B,Cgroup along with high fat and high sucrose diet,oral with L-Arabinose,the dosage were 250mg/d,150mg/d and 30mg/d respectively for 4 weeks.The body weight and the amount of food intake were monitored every 5 day and every 2 day,respectively,for 4 weeks,the fasting glucose,sucrose tolerance were examined using commercial glucose strips (Lifescan,One Touch Ultra,Melitas,CA).after 30 day experiment,all the mice were sacrificed and morphological examination on their liverswere performed.and PAS stained.Results:The experimental result proved that the L-Arabinose decreased significantly sucrose absorption,lower the fasting glucose level and the data of aera-under-curve (AUC)significantly in the mice with high fat and high sucrose fedding.The high dosage and middle dosage group,the L-Arabinose can decrease the weight increse,the structure of liver cell in D group mice and part of C group mice were damaged significantly and steatosis,PAS stained strong possitive,but in group A and group B,this kind of phenomenoe were not obvious.Conclusion: These results support the hypothesis that the L-Arabinose preparation by Thomson biotech(Xiamen)PTE LTD proved to be highly effective in preventing a rise in circulating glucose after sucrosechallenge,The body weight gain was less in the L-Arabinose treated group from 5th day of than that in the control group,the AUC data were significantly lower in L-Arabinose group than that of control group.the L-arabinose can inhibit the absorted of sucrose which usually be used for lipogenesis,the adipose in liver cell and other tissue was also decreased.
