摘  要:研究内生青霉菌(Penicilliumsp.B-4)胞外纤维素酶在槐米总黄酮提取中的辅助应用。内生青霉菌在起始pH4.5的综合马铃薯培养基中,150 r/min,40℃下摇瓶,培养7 d,具有较高的纤维素酶比活力(3.57 U/mL)。槐米干粉投入青霉菌发酵液中进行酶解处理,比较酶料比、酶解温度、酶解时间和酶解液pH对槐米总黄酮提取率的影响,发现槐米干粉以酶料比40∶1(mL/g)加入粗酶液中,在pH4.5、温度40℃下酶解处理1 h后,黄酮提取率可达12.2%,比常规提取率增加了38.7%。内生菌纤维素酶辅助提取法为槐米黄酮提取的可行新方法。The extracellular cellulase activity of an endophytic Penicillium sp. B-4 from Artemisa annua reached 3.57 U/mL when the fungus was cultivated in revised PDA medium with initial pH 4.5 at 40℃ on a rotary shaker incubator at 150 r/min for 7 d. The crude cellulase from the cultural medium was applied for the flavonoid extraction from flower buds of Sophara japonica. The ratio of dry flower bud to crude enzyme liquid, temperature,time, and pH value was optimizated during the process of cellulase-assisted treatment. After incubation with 40:1 of crude enzyme (pH 7.0) to dry flower bud for 1 h,the extraction rate of total flavonoids was 12.2% , and increased by 38.7% than that in regular extraction process. Results showed that the cellulase-aided extraction process on total flavones of Sophara japonica is practical.
