摘 要:目的研究去甲基化药物5-氮杂胞苷(5-Aza-CdR)对人结肠癌HT29细胞株生物学行为及肝癌缺失基因1(DLC-1)表达的影响。方法5-Aza-CdR处理HT29细胞72 h;RT-PCR检测DLC-1 mRNA的表达;MTT、平板克隆实验、Transwell小室实验分别检测药物处理前后的HT29细胞增殖、侵袭、迁移能力的变化。结果经5-Aza-CdR作用后,HT29细胞DLC-1恢复表达,细胞增殖活性降低,侵袭能力与迁移能力下降。结论5-Aza-CdR可抑制HT29细胞增殖、侵袭、迁移,可能与DLC-1基因恢复表达有关。Objective To observe the effects of 5-Aza-2'-deoxycytidine (5-Aza-CdR) on the proliferation, invasion, migration ability and frequently deleted in liver cancer-1 (DLC-1) gene expression of HT29 cell line. Methods The human colon cancer HT29 cell line was treated by various concertrations (5,10 μmol/L) of 5-Aza-CdR for 72 h. DLC-1 mRNA expression profile was studied by RT-PCR in pre- and post-treated cells. MTT assay was used for the test of cell proliferation The invasion and mobility abilities of the cells in vitro were explored by colony-forming experiment and transwell chamber. Results Compared to the cell lines without treatment, the proliferation of HT29 cell lines cultivated with 5-Aza-CdR was decreased and the invasion and mobility abilities of the cells in vitro were lowered. The expression level of DLC-1 mRNA in HT29 was restored after treated by 5-Aza-CdR. Conclusion 5-Aza-CdR inhibits the malignant biological behaviors of HT29 cells,which may be related to the restoration of DLC-1 gene expression.
