摘 要:目的鉴定乳腺癌组织、癌旁组织、正常组织与MCF-7细胞系中SP(side population)亚群含量是否有差异.方法制备乳腺癌组织、癌旁组织、正常组织细胞悬液和MCF-7细胞悬液,一组加入Hoechst33342染色,另一组同时加入拮抗剂维拉帕米,通过流式细胞仪测定SP亚群含量.结果SP亚群在癌组织、癌旁组织、正常组织和MCF-7细胞的含量分别为(0.54±0.17)%、(7.23±1.12)%、(8.34±0.96)%和(9.58±1.27)%,且它们之间的SP亚群含量具有差异显著性.结论人乳腺癌组织、癌旁组织、正常组织与MCF-7细胞系中都存在SP亚群且含量不同,能够为肿瘤干细胞的鉴定和提纯提供一定的科学参考价值.Objective To identify whether the percentages of SP cells have differences in breast cancer tissues, tumor-adjacent tissues, normal tissues and MCF-7 cancer cells. Methods After staining with Hoechst 3 3342 dye and Verapamil, the percentages of SP cells were detected in cell suspension from breast cancer tissues, tumor-adjacent tissues, normal tissues and MCF-7 cancer cell line respectively with the FACS method. Results The percentages of SP cells in breast cancer tissues, tumor-adjacent tissues, normal tissues and MCF-7 cancer cell line were (0.54 + 0.17)%, (7.23 + 1.12)%, (8.34 + 0.96)% and (9.58 + 1.27)%, respectively. There were significant differences between them. Conclusions Side population cells express in breast cancer tissues, tumor-adjacent tissues, normal tissues and MCF-7 cancer cell line, and the percentages of SP cells are different. This result can supply the scientific value to identify and purify breast cancer stem cells.
