摘  要:目的:研究散发性阿尔茨海默病(AD)患者脑内sorLA蛋白表达水平,并探讨其与脑内淀粉样斑块数量的关系。方法:随机选择15例尸检明确诊断的散发性AD患者(AD组)和15例非AD患者(对照组),采用Western-blot测定2组颞叶皮质组织中sorLA蛋白表达水平,并分析AD组脑内sorLA水平与淀粉样斑块数量的关系。结果:2组性别、年龄、死后尸检时间等匹配。AD组颞叶皮质内sorLA蛋白表达水平较对照组下降达63%(P<0.05),线性回归分析显示AD组脑内sorLA水平与淀粉样斑块数量明显呈反比。结论:散发性AD患者脑内sorLA蛋白表达显著下降,且其表达量与淀粉样β蛋白(Aβ)的沉积量直接相关,提示sorLA可能涉及散发性AD的发病机制。Objective: To determine the levels of sorLA in the brains of Alzheimer's Disease fAD) patients, and to observe the correlation of sorLA levels and plaques and beta-amyloid (Aβ) loads in AD brains. Methods: We have randomly selected 15 autopsy-confirmed sporadic AD and 15 control individuals, and quantitatively analyzed protein levels of sorLA in the temporal cortex with Western-blotting analysis and its association with plaque numbers in AD brains has been explored. Results: Patients having sporadic AD expressed significantly less sorLA protein in the temporal cortex when compared with controls. SorLA was decreased in the sporadic AD cases by 63% (P(0.05). By using linear regression analysis, we observed significant inverse correlations between levels of sorLA and plaque numbers in AD brains. Conclusion: The results of the current study suggest that sorLA loss might be an important factor in AD pathogenesis.
