摘 要:目的探讨趋化因子受体CXCR4在人乳腺癌中的表达及其与乳腺癌预后相关因素的关系。方法收集2004年9月-2006年1月手术切除的乳腺癌标本44例,采用RT-PCR检测CXCR4mRNA的表达,Western blotting检测CXCR4蛋白的表达,免疫组化检测CXCR4、雌激素受体(ER)、孕激素受体(PR)及人类表皮生长因子受体2(HER2,又称C-erbB-2)的表达,并观察相应的临床病理指标如年龄、月经状况、淋巴结转移数目、肿瘤大小、组织学分级等。结果免疫组化显示CXCR4主要在胞质表达,很少在细胞核表达。44例乳腺癌标本中均有不同程度的CXCR4mRNA及蛋白表达,且其表达水平随腋窝淋巴结转移增多而增高(P<0.05)。与腋窝淋巴结转移阴性者比较,腋窝淋巴结转移≥4个及1~3个者CXCR4mRNA及蛋白表达水平显著增高(P<0.05)。C-erbB-2(/)组与C-erbB-2(-/+)组比较,CXCR4mRNA及蛋白表达水平亦显著增高(P<0.05)。CXCR4mRNA及蛋白表达水平与患者年龄、月经状况、肿瘤大小、组织学分级、ER、PR无相关性(P>0.05)。结论CXCR4可能对促进乳腺癌转移有重要作用。Objective To study the expression of chemokine receptor-4 (CXCR4) in human breast cancer and its correlation with the prognostic factors. Methods Forty four samples of breast cancer were collected from Sep. 2004 to Jan. 2006. The expressions of CXCR4 mRNA and protein were evaluated by RT-PCR and Western blotting respectively. The expressions of CXCR4, estrogen receptor. (ER), progesterone receptor (PR) and GerbB-2 were examined by immunohistochemical methods. The clinicopathological features, such as ages, menstrual status, lymph node metastasis, tumor size and histological grading were recorded and analyzed. The relationship between CXCR4 and the clinicopathological features was analyzed. Results The immunohistochemical staining showed that CXCR4 expressed mainly in cytoplasm and little in nuclei. CXCR4 mRNA and protein expressed differently in 44 samples of breast cancer, and the expressive level in creased with the increase in metastatic lymph nodes number. Compared with axillary lymph nodes metastasis negative group, the expression of CXCR4 mRNA and protein increased significantly in ≥4 and 1-3 nodes metastasis groups (P〈0.05). Compared with C-erbB-2 (-/ +) group, the expression of CXCR4 mRNA and protein increased significantly in C erbB-2 (++/+++) group (P〈0. 05). However, no sig nificant correlation existed between the expression of CXCR4 mRNA and protein and the clinicopathological features such as ages, menstrual status, tumor size, histological grading and ER and PR (P〉0. 05). Conclusion CXCR4 might play an important role in promoting the metastasis of breast cancer.
