摘 要:目的研究三阴性乳腺癌(TNBC)中脂肪酸合成酶(FAS)及血管内皮生长因子-C(VEGF-C)的表达及临床意义。方法采用EnVision法从蛋白水平检测30例TNBC及25例非三阴性乳腺癌(non-TN-BC)、10例癌旁正常乳腺组织中FAS及VEGF-C的表达情况。观察TNBC中FAS及VEGF-C表达及与临床病理特点之间的关系。结果 TNBC中FAS的阳性表达率[60%(18/30)]明显高于non-TNBC[24%(6/25)];VEGF-C的阳性表达率[30%(9/30)]明显低于non-TNBC[80%(20/25)];TNBC中FAS表达与患者的年龄、月经状态、分期、淋巴结状态无关(P>0.05),而与肿瘤大小、远处转移有关(P<0.05)。肿瘤大、疾病进展快的患者,FAS表达较肿瘤小、进展慢的患者强,而VEGF-C在TNBC中的表达与患者年龄、月经状态、肿瘤分期、肿瘤大小和淋巴结累及情况均无关(P>0.05)。结论 TNBC中FAS的高表达可能在促进肿瘤生长、转移中起作用,FAS可能成为TNBC治疗的一个分子靶点,而针对VEGF-C蛋白靶向治疗可能更适合non-TNBC。Objective To detect the expression of fatty acid synthase(FAS) and vascular endothelial growth factor-C(VEGF-C) in triple negative breast cancer and its significance.Methods The expression of FAS and VEGF-C in triple negative breast cancer was detected using immunehisto-chemistry.The correlation between the FAS and VEGF-C expression and the clinical pathological characteristics of triple negative breast cancer was observed.Results FAS over expressed in TNBC,expressed at high levels(60%),but low expressed(24%)in non-TNBC.VEGF-C over expressed in non-TNBC,expressed at high levels(80%),but low expressed(30%)in TNBC.There were significant differences of FAS and VEGF-C expression between TNBC and non-TNBC(P<0.05).The expression of FAS was not correlated with patients age,menopausal status,stage,lymph node status(P>0.05),but was correlated with the tumor size,distant metastasis(P<0.05).The expression of FAS in patients with large tumor,progress fast was stronger than in patients with small tumor,progress slow.The expression of VEGF-C in TNBC was not correlated with patients age,menopausal status,stage,tumor size,lymph node status(P>0.05).Conclusions FAS may play a role in tumor growth,metastasis in triple negative breast cancer,FAS might serve as a biomarker for the treatment.VEGF-C serve as a biomarker for the treatment is more fitter in non-triple negative breast cancer.
