摘 要:参照糖单孢菌属(Saccharomonospora)和拟诺卡氏菌属(Nocardiopsis)的特异性引物,建立多重PCR反应体系快速鉴定分离自新疆艾丁湖盐碱环境分离的118株嗜盐菌株,检测出14株糖单孢菌属菌株,17株拟诺卡氏菌属菌株.研究发现2株普氏菌属菌株出现针对糖单孢菌属的特异性条带,退火温度提高至61℃时可消除这种假阳性扩增.应用多重PCR快速鉴定方法可提高对盐碱环境中的这2个类群初步鉴定以及筛选工作的速度和效率.A genus- specific PCR method described previously was used for identification of novel strains of the genera Saccharomonospora and Nocardiopsis that isolated from the soil sample collected from Aiding Lake in Xinjiang, China. 118 strains were used in this research and 14 strains of Saccharomonospora and 17 strains of Nocardiopsis were detected. In this process, two Prauserella strains were also amplified by using Saccharomonospora - specific primers. However, when PCR annealing temperature was adjusted to 61 ℃ ,this pseudo- positive amplification could be removed. In order to shorten the process of identification and screening, a stable multiplex PCR system that can be applied to make the detection of these two genera simulataneously was established.
