解毒维康片对HL-60白血病细胞血管内皮生长因子表达的影响

摘　　要：目的探讨中药复方制剂解毒维康片(JDWKP)对HL-60白血病细胞血管内皮生长因子(VEGF)表达的影响。方法采用血清药理学方法,以大鼠为实验动物,制备出JDWKP的含药血清,再采用酶联免疫吸附法(ELISA法),检测含药血清作用于HL-60细胞不同时间后培养上清中VEGF的含量。结果JDWKP含药血清能以剂量依赖方式明显抑制HL-60细胞VEGF的表达,中等剂量含药血清处理HL-60细胞48 h,VEGF的浓度仅为阴性对照组的21.6%,但进一步增高药物浓度或延长作用时间,VEGF浓度并无明显的相应改变。结论JDWKP能下调HL-60细胞VEGF的表达水平,这可能是中药复方制剂JDWKP抗白血病的作用机制之一。Objective To investigate effects of Chinese herbs mixture jieduweikang pill (JDWKP) on the expression of vascular endothelai growth factor (VEGF) derived from leukemia cell line HL-60.Methods The serum containing the JDWKP was collected from rats by using the method of pharmacology of the serum. Enzyme linked immunosorbent assay (ELISA) was used to detect the expression of VEGF in the cultural su- pematant of HL-60 cells treating with JDWKP.Results The expression of VEGF was evidendy downregulat- ed in a concentration-dependent manner after treating with the serum containing JDWKP and the concentra- tions of VEGF were only 21.6 % compared with the negative control after 48 hours treating with JDWKP. Conclusion The expression of VEGF derived from leukemia cell line HL-60 could be downregulated by the serum-containing JDWKP,which may be one of the mechanisms of antileukemia effects exerted by JDWKP.
