环氧化酶-2在子宫内膜样腺癌中的表达及其与PTEN的相关性

摘　　要：目的研究环氧化酶-2(COX-2)在子宫内膜样腺癌(EAC)中的表达及意义,探讨COX-2与PTEN的相关性。方法用免疫组化SP法检测COX-2、PTEN在正常子宫内膜、子宫内膜非典型增生及EAC中的表达。结果COX-2的阳性表达率自正常内膜至非典型增生及EAC呈阶梯性升高(13.33%、50.00%、64.58%)(P<0.05)。EAC中,COX-2蛋白表达与组织分级(G_1、C_2、G_3分别为4/9、64.29%、81.82%)、肌层浸润(深肌层浸润、浅肌层浸润分别为75.00%、57.14%)、手术-病理分期(早期、晚期分别为56.41%、9/9)、淋巴转移(有、无分别为9/9、56.41%)相关(P=0.007,P=0.009.P= 0.001,P=0.001)。预后好的EAC中,COX-2与PTEN之间存在负相关(r_(?)=-0.400,P<0.05)。结论COX-2与EAC的浸润转移相关;预后好的EAC中,PTEN可能参与COX-2的表达调控。Objective To study the expression and clinical significance of COX-2 and the relation- ship between COX-2 and PTEN in endometrial adenocarcinoma(EAC).Methods The expression of COX-2, PTEN protein was detected by SP-immunohistochemical method in EAC,endometrial intraepithelial neoplasia, corresponding normal endometrium.Results The positive rates of COX-2 protein increased from normal en- dometrium(13.33%) to endometrial intraepithelial neoplasia(50.00%) and adenocarcinoma(64.58 %)(P<0.05).COX-2 positive ratio was higher in poorer differentiation (81.82% vs 64.29% vs 4/9,P=0.007), deeper myometrial invasion(75.00% vs 57.14%,P=0.009),extrauterine invasion/metastasis and later stage (9/9vs 56.41%,P=0.001)(P<0.05).In EAC with favorable prognosis,the expression pattern of COX-2 was correlated with PTEN(r_s=-0.400,P<0.05).Conclusion COX-2 may be a parameter indicating tumor ag- gressiveness.In EAC with favorable prognosis,PTEN may regulate the expression of COX-2 protein.
