摘 要:目的比较不同浓度自体骨髓基质细胞修复牙周组织缺损的能力。方法在6只一岁半的Beagle犬的下颌牙制备慢性Ⅱ°根分叉病变模型,在GTR的基础上,用不同浓度的BMSCs(5×105/ml组,5×106/ml组,5×107/ml组)移植治疗,12周后作病理切片,HE染色观察并测量牙周组织再生情况。结果GTR组的新生牙骨质长度和新生牙槽骨面积的百分比分别为:51.5±5.6和27.1±7.7;5×105/ml组为:84.8±8.9和30.6±7.7;5×106/ml组为:91.8±5.2和68.3±11.4;5×107/ml组为:88.8±7.2和78.5±12.7。细胞组的新生牙骨质量与对照组相比差异有显著性(p<0.01),但各细胞组间相互比较无差异显著性;5×106/ml组和5×107/ml组的新生牙槽骨量与5×105/m/组和对照组相比差异有显著性(p<0.01),但前两组间和后两组间比较则差异无显著性。结论骨髓基质细胞促进牙周组织再生的作用与细胞的接种密度有关,高浓度细胞能有效促进牙周组织再生。Objective To compare the difference in promoting periodontal regeneration of initial cell seeding density in canine experimental class II furcation defects . Methods GTR combination with the expanded BMSCs which were seeded in collagen membrance at initial concentrations of 5×105 cells/ml,5×106 cells /ml or 5×107 cells /ml, and auto-transplanted into experimental class II furcation defects, underneath a Bio-Gide membrane. Bio-Gide membrane alone was implanted into the defects as a control. Periodontal regeneration was evaluated by histologic and histometric analyses 12 weeks after transplantation. Results The percentage of new cementum length in the 5×105 cells/ml,5×106 cells /ml and 5×107 cells /ml groups were significantly higher than in the control group (p<0.01), and there was no significantly difference in the three cell groups. The percentage of new alveolar bone area in the 5×106 cells /ml and 5×107 cells /ml groups were significantly higher than in the control and 5×105 cells/ml group (p<0.01), and there was no significantly difference between the first two groups, and neither was the later. Conclusions This study suggested that the effect of auto-transplanted BMSCs promoting periodontal healing has cell density-dependent under certain conditions, and the higher concentrations(5×106 cells /ml and 5×107 cells /ml) can effectively promote periodontal regeneration.
