摘  要:冷冻干燥是制备蛋白质药物常用方法.详细综述了冷冻干燥过程中导致蛋白质变性机理的种类、特征、原理;介绍了主要的保护措施即添加保护剂,并阐述了保护剂的作用机理:优先化作用、玻璃态假说和水代替假说.The method most commonly used for pharmaceutical protein products is lyophilization. The classification, characters, and mechanism of denaturation protein on Freeze-dry process are thorough discussed. To protect a protein from freezing and dehydration, a protein stabilizers may be used. There are primary three mechamism of protein stabilization during lyophilization, which are the preferential interaction, the fonnation of an amorphous glass and the water replacement.
