摘  要:目的观察知母皂甙元对青年和老年大鼠脑M及M1受体的不同影响。 方法以对M1受体有相对选择性的哌仑西平为非标记竞争剂 ,3H -QNB为非选择性标记配基 ,建立M和M1受体的放射配基竞争结合分析方法 ,观察知母皂甙元对青年大鼠和老年大鼠脑M和M1受体的影响。 结果与青年大鼠相比 ,老年大鼠脑M及M1受体减少 ,知母皂甙元能上调M和M1受体 ;对正常青年大鼠脑M及M1受体的密度 ,知母皂甙元没有影响。结论知母皂甙元对下降的M及M1受体密度有明显的上调作用 ,但不超过正常值 ,对正常水平的M及M1受体密度 。Objective To explore whether the effects of a sapogenin from Zhimu (ZMS) on cerebral M and M 1 receptor densities are different in young rats and aged rats. Methods A dual site competitive binding technique was established,using unlabelled pirenzipine as the selective competitive agent and 3H-QNB as the nonselective radioligand. This technique was used to observe the total cerebral M and M 1 receptor densities in aged and young SD rats, treated or untreated with ZMS. ResultsIn aged rats, the densities of total cerebral M and M 1 receptors were both significantly decreased. Oral administration of ZMS for three months up-regulated the total cerebral M and M 1 receptors in aged rats, making the density approaching normal, but similar administration of ZMS has no effect in young rats. Conclusion ZMS can only restore the decreased total cerebral M and M 1 receptor densities, making them approaching normal, but cannot raise the normal densities to a higher level.
