摘 要:为了解p16基因在急性髓系白血病(AML)发病中的意义,用PCR-SSCP和Northern blot方法分析了16例AML细胞p16基因结构及mRNA的表达。研究结果表明,16例未发现p16基因的缺失、突变。初治和复发的12例AML的p16 mRNA表达水平明显低于4例长期缓解者及正常对照者。1例p53突变AML的p16 mRNA有明显升高,提示p16基因在AML发病中可能不占重要地位,而在AML细胞抑制正常造血的逐步恶化中起一定作用,在p53突变时,其表达反馈性升高显示一种反馈环路失衡。To detect the role of p16 gene in the pathogenesis of AML, PCR-SSCP and Northern blot were used respectively to analyse the structure and mRNA expression of p16 gene in 16 cases of AML cells. No depletion or mutation of p16 gene were found in all the 16 cases. The p16 mRNA expression in 12 initial and recurrent cases was significantly lower than that in 4 cases with long term remission and normal control. The p16 mRNA expression in 1 case of AML with p53 mutation was enhanced obviously. p16 gene may not have an important role in the pathogenesis of AML, while may have certain inhibition effects on the progressive deterioration of normal hematopoiesis in AML cells. The feedback rise of p16 expression in the case with p53 mutation showed a kind of imbalance of the feedback cycle.
