提取百日咳保护性抗原的简易方法

摘　　要：用蓝色琼脂糖 CL—6B 做亲和吸附凝胶,仅用一步简单操作即可将1相百日咳菌在液体培养基中产生的 FHA 和 PT 抗原全部提纯出来。用 cs 菌株和 Tohama 菌株所得的提取物中,具有全部 FHA 和 PT 的生物学活性,共总抗原提取量二者没有区别,但 Tohama 菌株提取物的毒性略高于 cs 菌株。提取物中内毒素含量极微,未查出有凝集原存在。本方法为简化无细胞百日咳菌苗的研制过程提供了有利条件。A purfied extract which containing two kinds of protective antigens Filamentous Hemagglutinin(FHA)and Pertussis toxin(PT)could be obtained from culture supern- atant of phase ⅠB.pertussis by a simple procedure using Blue-Sepharose Cl-6B chrom- atography.This extract possessed all biological activities of FHA and PT.The total amount of extracted antigens were found no difference between CS strain and Tohama strain,but the toxic activities were higher in Tohama strain than that of CS strain.The LPS was rare and agglutinogens could not be detected in the extract. This paper provided a useful method for the production of acellular pertussis vaccine.
