测定血清硒含量的简易消化法

摘　　要：本文提出测定血清硒的简易消化方法。以硝酸—高氯酸作消化液,用普通 10ml 具磨口玻璃塞试管作消化器在电热套砂浴上消化。取样量仅需100μl,消化过程耗时在2小时以内。用荧光法测定消化液硒含量,方法相对均方偏差为2.2％,回收率97％,全部过程耗时不超过半天。样品在4℃冰箱内保存数日后复测,其荧光强度与第一次测定值之间相对偏差不超过3％。该方法特点是对试剂、器皿及设备没有特别严格的要求,因而简捷、实用。适于测定血液及生物组织中微量硒。In this paper, a simple wet digestion method for determination ofselenium in serum was presented. A 0.1ml sample was placed into a10ml test tube with a glass stopper was digested with a mixture ofHNO-HC1 on a sand-bath heating mantle. The digestion takes twohours, the selenium content in the sample was determined by fluorescencespectrophotometry. The CV% of determination is 2.2%; the recoveryis 98%. After keeping in a refregeritor at 4℃ for a few days, the samesample was determined again. The repeation of mean value in eachredetermination is less than 3%. This method can also be used fordetermination of selenium in other biological samples other than serum.
